要約

腫瘍抑制因子PTENは、そのホスファチジルイノシトール-3,4,5-三リン酸[PI(3,4,5)P3]ホスファターゼ活性が主にホスホイノシチド3キナーゼ(PI3K)の発がん能を直接的に相殺することにより、細胞形質転換の主要なブレーキとなっている。PTENの変異は、腫瘍で、および腫瘍の素因を有する患者や神経障害または認知障害を有する患者の生殖系列で高頻度に認められることから、PTENの遺伝子とタンパク質は現在の生物医学研究において大きな関心の的になっている。403残基長のPTENタンパク質に関する約20年の徹底した調査の末に、別の翻訳開始部位から生じた結果としてアミノ末端に173アミノ酸からなる伸長部をもつ、それまで特徴づけられていなかった形態のPTENが発見されている。この分野の研究を促進し、論文およびデータベースに記載されるPTENアミノ酸の命名と識別における曖昧性を回避するために、われわれは、この長鎖型のPTENのための統一命名法とアミノ酸位置番号を提唱する。
