要約:動的予測モデルは、複雑な生物システムの解析に重要である。しかし、システム生物学モデルを体系的に開発して識別する方法は、まだ十分でない。われわれは、生物システムの構造に関するすべての仮説的機構を単一のモデルに組み入れ、よりシンプルで観察データと適合するモデルセットを自動生成する計算方法を記述する。原理の証明として、われわれは出芽酵母(Saccharomyces cerevisiae)の転写因子Msn2の動的制御、とくに飢餓ストレスから解放されたあとの細胞の回復に関わる短期的機構を解析した。われわれの方法によって、考えうる192のモデルのうち12が、利用可能なMsn2の局在化データと適合すると決定された。モデル予測と合理的にデザインされたホスホプロテオミクスと画像化実験の繰り返しによって、192のモデルのなかから相対的確率99%の単一回路トポロジーが同定された。モデル解析では、Msn2のリン酸化と輸送における動的現象の共役が変化率センサーとして確立されることによって、効率的なストレス反応シグナル伝達が引き起こされうることが示された。同様の原理は、哺乳類のストレス反応経路にも当てはまる可能性がある。動的モデルの系統的構成が、明らかではない分子機構についての深い洞察を生むかもしれない。
