要約 成長中の血管や損傷した血管において、血管平滑筋細胞(VSMC)は、転写段階で調節される可逆的分化を受ける。この脱分化は、血管形成術やステント留置術に起因する損傷などの、血管損傷後のVSMC過形成にも関与する。ステントは、機械的支持を提供するとともに、機構的ラパマイシン標的タンパク質複合体1(mTORC1)の阻害物質であるラパマイシンを含有し、放出することがある。ラパマイシンは、VSMC過形成を抑制し、VSMC分化を促進する。われわれは、VSMCのラパマイシン誘導性分化には転写因子GATA-6が必要であったことを報告する。mTORC1阻害によって、GATA-6が安定化し、GATA-6の核内蓄積、DNAとの結合、収縮タンパク質をコードするプロモーターの転写活性化、増殖の抑制が促進された。これらの作用は、mTORC1阻害時にVSMCで活性化されるキナーゼAkt2によると考えられる、GATA-6のSer290でのリン酸化を介して生じた。ラパマイシンは、野生型マウスではGATA-6のリン酸化を誘導したが、Akt2−/−マウスでは誘導しなかった。動脈損傷後の内膜過形成は、Akt2−/−マウスにおいて野生型マウスよりも大きく、Akt2−/−マウスにおける反応の悪化は、野生型GATA-6またはリン酸化模倣(S290D)変異GATA-6の局所過剰発現によって、リン酸化欠損(S290A)変異体を局所過剰発現させた場合よりも、大きくレスキューされた。われわれのデータからは、GATA-6とAkt2が、VSMCの増殖および分化のmTORC1を介する調節に関与することが示された。mTORC1の下流転写標的を同定することで、VSMCの過剰増殖を伴う心血管疾患と闘うための、細胞種特異的な薬剤標的がもたらされる可能性がある。
