要約 巨核芽球性白血病1(MKL1)は、細胞増殖、運動性、接着性、および分化など細胞内の動的な細胞骨格変化も伴う過程に関連する遺伝子の発現を促進する血清応答因子(SRF)のコアクチベーターである。MKL1は、単量体球状アクチン(G-アクチン)に結合していると不活性であるが、低分子量グアノシントリホスファターゼRhoAを活性化させるシグナルは、アクチンの重合とG-アクチンからのMKL1の解離を引き起こす。われわれは、繊維状アクチン(F-アクチン)に結合するタンパク質、フィラミンA(FLNA)との結合により媒介されるMKL1の新しい活性化機構を発見した。FLNAとMKL1の相互作用は、初代線維芽細胞、メラノーマ、乳癌および肝細胞癌細胞においてMKL1の標的遺伝子の発現に必要であった。われわれは、MKL1とFLNAの相互作用領域を同定し、FLNAと結合できないMKL1の変異体を発現する細胞では、細胞遊走が損なわれ、MKL1-SRFの標的遺伝子の発現が低下した。MKL1-SRFの標的遺伝子の誘導および抑制はそれぞれ、MKL1-FLNAの相互作用の増加あるいは減少と相関していた。初代ヒト線維芽細胞におけるリゾホスファチジン酸誘導性のRhoAの活性化は、FLNAと内因性MKL1の会合を促進したのに対し、アクチン重合阻害剤への曝露は、メラノーマ細胞においてFLNAからMKL1を解離させ、MKL1-SRFの標的遺伝子の発現を減少させた。このように、FLNAは、アクチンの重合とMKL1-SRFの活性を関連づけ、MKL1と単量体G-アクチンの既知の抑制複合体に対抗する正の細胞トランスデューサーとして機能する。
