ケモカインは、細胞表面ヘテロ三量体グアニンヌクレオチド結合タンパク質(Gタンパク質)共役受容体(GPCR)に結合することによって細胞遊走を組織化し、発生、免疫監視、疾患に寄与する。ケモカイン約50種類とその標的20種類の多様な相互作用によって広範なシグナル伝達ネットワークが生成されており、そこに複数のアゴニストと結合できる性質とそのバイアスが合わさって一層複雑化している。受容体CXCR4は、ケモカインCXCL12の単量体と二量体の両方を認識し、ケモカインファミリーのリガンドバイアスの特徴をよく示している。われわれは、恒常的に単量体となるCXCL12変異体によって、正常なCXCR4シグナル伝達に伴うGタンパク質依存性の応答とβアレスチン依存性の応答が再現され、細胞遊走が引き起こされることを示した。さらに、CXCL12単量体とCXCR4との間には、Gタンパク質依存性シグナル伝達のみを亢進するバイアス型アゴニストであるCXCL12二量体と複合体を形成した受容体の構造にはみられない特異的な接触がみられた。われわれは、CXCR4のアミノ末端と結合したCXCL12単量体の核磁気共鳴に基づく構造をCXCR4の膜貫通ドメインの結晶構造と重ね合わせ、実験的に検証されたアゴニスト結合型ケモカイン受容体のモデルを作製した。このモデル構造によって明らかにされた巨大なCXCL12:CXCR4タンパク質間境界面から、これまで特徴づけられていない機能的相互作用がケモカイン受容体認識の古典的な「2部位モデル」の範囲外に同定された。われわれのモデルは、CXCR4受容体の細胞外面での相互作用がケモカイン受容体に媒介されるシグナル伝達に必要な立体構造変化を促進しているとする機構的な仮説を示唆している。
