アダプタータンパク質LAT(活性化T細胞のリンカー)を中心とするシグナルソームの正確な構築は、T細胞およびマスト細胞の抗原受容体シグナル伝達に必要である。シグナルソームの構築時、細胞質アダプタータンパク質の増殖因子受容体結合タンパク質2(Grb2)ファミリーのメンバーは、リン酸化チロシン(pTyr)残基との相互作用を介してLATと協同的に結合する。われわれは、Grb2ファミリーメンバーであるShc下流Grb2関連アダプター(Gads)のSrc相同性2(SH2)ドメインを介した二量体化を示した。Gadsの二量体化はSH2ドメイン境界面を介してなされるが、この面はpTyr結合ポケットとは別の場所であり、対になったGadsのLATへの協同的、優先的結合を促進した。数理モデルにより定量化された、このSH2ドメインに内在的な協同的機構は、LAT分子の一重、二重リン酸化状態をGadsが区別することを可能にした。二量体化境界面の突然変異による不活性化は、協同性を低下させ、T細胞およびマスト細胞におけるGadsシグナル伝達を妨げた。Gadsの二量体化依存的、協同的なLATへの結合は、不完全にリン酸化されたLAT分子とのシグナルソーム形成を減少させることにより抗原受容体の感受性を増加させ、それにより完全なシグナルソームの形成に優先させる可能性がある。
