要約:キナーゼの哺乳類ラパマイシン標的タンパク質(mTOR)は、2つの多タンパク質複合体(mTORC1、mTORC2)として存在し、増殖と代謝の中心的な調節因子である。ホスホイノシチド3-キナーゼ(PI3K)、Akt、抑制性の結節性硬化症1/2(TSC1-TSC2)を介するmTORC1のインスリンによる活性化は、負のフィードバックループを始動させ、最終的にPI3Kを抑制する。今回われわれは、コアネットワーク全体を統合するデータ駆動型の動的インスリン- mTORネットワークモデルについて紹介、このモデルを用いて、十分にはわかっていないインスリンによるmTORC2の調節機構について検討した。われわれは、in silicoと実験において、ネットワーク内のいくつかのレベルを標的とする撹乱の影響を解析することによって、現在の仮説とは異なり、TSC1-TSC2複合体はmTORC2の直接的な調節因子でも間接的な調節因子(負のフィードバックループを介して作用する)でもないことを見いだした。mTORC2の活性化には活性型のPI3Kが必要であったが、これは負のフィードバックループによる影響を受けなかった。そこでわれわれは、mTORC1を調節する負のフィードバックループに非感受性のPI3Kのバリアントを介するmTORC2の活性化経路を提唱する。この推定上の経路から、mTORC2はTSC1-TSC2を抑制するAktに応答しないことが予測され、実際にわれわれは、mTORC2がいくつかの細胞種において、Aktの構成的活性化に対して非感受性であることを見いだした。われわれの結果は、これまで知られていなかったネットワーク構造が、mTORC2をその上流のシグナルに結び付けることを示唆しており、mTORC2活性化に寄与する分子結合因子を明らかにしている。
