Background

Increasing evidence points to the adverse effects of advanced glycation end product modified proteins on aging-related human health. AGEs modified proteins result from the non-enzymatic glycosylation of glucose with free protein amino groups reaction of carbohydrates via Maillard reaction, including advanced product formation following Amadori rearrangements. AGEs accumulation is thought to contribute to pathological changes resulting in cataract formation, Alzheimer‘s disease, osteoarthritis, and myocardial dysfunction. Diabetes (hyperglycemia) is associated with accelerated of AGE formation.
Result of a anti AGEs antibody analysis, shows that AGEs accumulates the following parts of human lens (nondiabetic and noncataractous), renal proximal tubules in patients with diabetic nephropathy and chronic renal failure, diabetic retina, peripheral nerves of diabetic neuropathy, atherosclerotic lesions of arterial walls, ß2-microglobulin forming amyloid fibrils in patients with hemodialysis-related amyloidosis, senile plaques of patients with Alzheimer disease, the peritoneum of CAPD patients, skin elastin in actinic elastosis, and ceriod/lipofuscin deposits. These evidence suggests that AGEs is deeply involved with Aging itself and chronic disease caused by an aging, and several AGE structure give emphasis to diabetes and brain disease field category. Various antibody Currently Proposed as AGE is CML,CEL, Pentosidine, Pyrraline, Imidazolone, Crossline and others.
On the basis of these findings,this kit was developed.

Measurement Principle

The Glucose-derived AGEs ELISA Kit is a solid phase enzyme-linked immunosorbent assay (ELISA) based on sandwich principle.

The microtiter wells are coated with a monoclonal antibody directed towards a unique antigenic site on Glucose-derived AGEs (Glc-AGEs).

When the sample diluted to an arbitrary density is added to well, Glc-AGEs is bonded.

In the use of HRP- anti AGE antibody, it is measured by the enzyme reaction of HRP.

Recovery test : In the recovery study, recoveries between 96% and 108% were obtained for 30 times dilutions of the sample sera (already-known AGEs conc. 300 ng/mL)
