Calcium-Binding Chaperones for Sperm Fertility



A large amount of cases of unexplained infertility in human males results from poor sperm quality. Calcium-binding chaperones such as Calsperin, Calmegin, PDILT and Calnexin are secondary messengers in the signal transduction pathway and play a major role in quality control by retaining incompletely folded or misfolded proteins in the endoplasmic reticulum (ER).

-defect in Calsperin gene leads to defective sperm migration and defective binding to the egg extracellular matrix (zona pellucida) 2

Calnexin/ CANX
-an integral membrane protein
-lectin chaperone located in the endoplasmic reticulum

-A testis-specific ER protein that is homologous to calnexin
-Specifically expressed during male meiotic germ cell development.
-Defect in Calmegin leads to a defect where sperm cannot adhere to egg extracellular matrix (zona pellucida)
-Functions as a chaperone for one or more sperm surface proteins that mediate the interactions between sperm and egg1

-Cooperates with the testis-specific calreticulin-like chaperone, calsperin (CALR3), in the endoplasmic reticulum
-Plays an indispensable role in the disulfide-bond formation and folding of ADAM3 (a gene which codes for a cysteine-rich transmembrane protein expressed in the testis).
-Pdilt(-/-) mice were male infertile because ADAM3 could not be folded properly and transported to the sperm surface without the PDILT/CALR3 complex.
