背景

ブロモドメインはエピジェネティックなリーダータンパクで、アセチル化ヒストンと結合し，クロマチンの構造や遺伝子発現を制御します。低分子阻害薬の開発において、疾患に関連性のあるブロモドメインを含むタンパク質は新たな創薬ターゲットとして注目されています。
BROMOscan™は低分子のブロモドメイン阻害剤を同定するための革新的な技術で、定量性のある活性部位依存的競合測定によりハイスループットスクリーニングから選択的プロファイリングに対応しています。

原理

DNAタグが付加されたブロモドメインは、活性部位にて固相化されたリガンドと結合します(Fig.1A)。
固相化リガンドと競合する試験化合物は、リガンド-ブロモドメイン間の結合を競合的に阻害し、リガンドと結合するタンパク量が減少します(Fig B)。ブロモドメインに結合しない試験化合物は、固相化されたリガンドに結合するブロモドメインの量に影響を与えません(Fig 1C)。
DNAタグを超高感度定量性PCR法（quantitate PCR; qPCR）により検出することで、リガンドに結合しているブロモドメインを定量化し、試験化合物と陽性対照薬剤間の比較をする事により、阻害活性の評価を行います(Fig 1D)。
試験化合物の濃度条件を振ることにより、試験化合物とブロモドメイン間の相互作用における解離定数（Kds）を測定することも可能です。
