使用目的

ジチオスレイトール（DTT）存在下で、PDI の触媒によりインスリンが還元されると、インスリンは凝集体を形成し、赤色蛍光体である PROTEOSTAT® PDI 検出色素に結合します。類似の比濁法による PDI 活性アッセイと比較して、蛍光ベースのアッセイは、シグナルウィンドウの大幅な向上、検出限界の改善、優れたZ'-スコア（>0.8）が特長となります。本アッセイのプレート内及びプレート間 CV は、通常 3-5% です。キットには、ヒトリコンビナント PDI 酵素（EC 5.3.4.1）、PDI 阻害剤であるバシトラシン、及び PDI 活性の変化をモニターするために必要な全ての試薬が含まれています。

PROTEOSTAT® PDI アッセイキットは、PDI 酵素活性をハイスループット形式で定量的に測定できるため、化合物ライブラリーからの PDI 阻害剤の同定にご使用いただけます。
