本キットは、遊離リン酸を検出する BIOMOL GREEN を組み合わせた、サイクリックヌクレオチド・ホスホエステラーゼ活性やその阻害剤、モジュレーターをスクリーニングするキットです。アッセイ原理は、サイクリックヌクレオチド・ホスホジエステラーゼがその基質である cAMP/cGMP を加水分解し、生じた 5'-ヌクレオチドをさらに 5'-ヌクレオチダーゼによりヌクレオシドとリン酸に開裂させることを利用しています。遊離したリン酸を BIOMOL GREEN™ で比色定量することで、酵素活性を測定できます。キットには、バリデーション用にI型cAMP ホスフォジエステラーゼ (PDE)、阻害剤スクリーニング用のテストコントロールとして非特異的なサイクリックヌクレオチド・ホスホジエステラーゼ阻害剤、3-isobutyl-1-methylxanthine (IBMX) が添付されています。本アッセイでは、スクリーニング化合物が 5'-ヌクレオチダーゼ活性を阻害しないことが重要になるので、阻害活性がないことを添付の 5'-AMP/GMP で確認することができます。

使用例

図1. PDEによるcAMP加水分解の継時変化（IBMX阻害の影響）
PDE酵素（20mU / well）を、阻害剤IBMX（40µM）の有無で、cAMP (200µM) 及び５'-nucleotidase (50kU / well) と共に、30℃下で指示された時間インキュベートした。反応はBIOMOL GREEN™ を100µL添加することで停止させ、その30分後にOD620を測定した。
