ヌクレオポリンタンパク質（Nucleoporin／NUP）抗体

各種ヌクレオポリンタンパク質（Nucleoporin／NUP）抗体を販売しております。

核膜孔複合体 (NPC) は高分子の核-細胞質間輸送における細胞核-細胞質間の橋渡しをする構造をしています。

NPCの主な構成要素であるヌクレオポリン（Nucleoporin／NUP）は約30種類あり、 FG-ヌクレオポリン、FC-ヌクレオポリン、そしてWD-リピートの7枚羽プロペラモチーフをもつヌクレオポリン、これら3ファミリーにクラス分けできます。ヌクレオポリンはサイズが多く、各分子量 (キロダルトン単位) によって名前がつけられています。NPC における各ヌクレオポリンの機能は重複せずに働きますが、その働きはまだ完全には分かっていません。分かっている機能として、8回の回転対称をもつ NPC を形成するために、サブ複合体をまとめる必須構造要因の働きをしています。また、NPC経由で高分子を能動輸送する際にも重要な役割をしています。しかし、モーターやATPase/GTPase のふるまいはみられません。

ヌクレオポリンの機能要因をもつ白血病誘発融合タンパク質の最近の研究では、転写におけるヌクレオポリンの新しい機能が見つかっています。これらの発見からヌクレオポリンの機能が広くバラエティに富んでいるということが分かります。

Detection of Human NUP153 by Western Blot and Immunoprecipitation.
Samples: Whole cell lysate from HeLa (5, 15 and 50 mcg for WB; 1 mg for IP, 20% of IP loaded) and 293T (T; 50 mcg) cells.
Antibody: Affi nity purifi ed rabbit anti-NUP153 antibody A301-788A used for WB at 0.04 mcg/ml (A) and 1 mcg/ml (B) and used for IP at 3 mcg/ mg lysate. NUP153 was also immunoprecipitated by rabbit anti-NUP153 antibody A301-789A, which recognizes a downstream epitope.
For blotting immunoprecipitated NUP153, the ReliaBLOTR Reagents and Procedures (Cat. No. WB120) were used. Detection: Chemiluminescence with exposure times of 3 minutes (A) and 1 second (B).

Detection of Human NUP50 by Western Blot and Immunoprecipitation.
Samples: Whole cell lysate (5, 15 and 50 mcg for WB; 1 mg for IP, 20% of IP loaded) from HeLa cells.
Antibody: Affi nity purifi ed rabbit anti-NUP50 antibody A301-782A used for WB at 0.1 mcg/ml (A) and 1 mcg/ml (B) and used for IP at 3 mcg/mg lysate. NUP50 was also immunoprecipitated by rabbit anti-NUP50 antibody A301- 783A, which recognizes a downstream epitope.
For blotting immunoprecipitated NUP50, the ReliaBLOTR Reagents and Procedures (Cat. No. WB120) were used.
Detection: Chemiluminescence with exposure times of 30 seconds (A)
and 3 seconds (B).

Detection of Human NUPL by Western Blot and Immunoprecipitation.
Samples: Whole cell lysate from HeLa (5, 15 and 50 mcg for WB; 1 mg for IP, 20% of IP loaded) and 293T (T; 50 mcg) cells.
Antibody: Affi nity purifi ed rabbit anti- NUPL antibody A301-792A used for WB at 0.04 mcg/ml (A) and 1 mcg/ml (B) and used for IP at 3 mcg/mg lysate.
NUPL was also immunoprecipitated by rabbit anti-NUPL antibody BL6839, which recognizes an upstream epitope.
For blotting immunoprecipitated NUPL, the ReliaBLOTR Reagents and Procedures (Cat. No. WB120) were used.
Detection:Chemiluminescence with exposure times of 10 seconds (A) and 3 seconds (B).
