背景

細胞内標的タンパク質に対する化合物の有効性評価

創薬研究において、リード化合物が細胞膜の透過性を持っているか、また、細胞内微小環境下において標的タンパク質とどの様に結合しているかを判定することは、リード化合物の構造探索と最適化の点において重要な意味を持っています。DiscoverX社のInCELL Hunter技術は、生細胞内における標的タンパク質の安定性の変化を検出することにより、Hit-to-LeadからLead Optimizationまで様々な研究用途において、リード化合物の細胞膜透過性を含めた標的タンパク質への結合能の評価を可能とします。

原理

本アッセイでは、DiscoverX社独自の技術；Enzyme Fragment Complementation (EFC)技術を応用し、細胞内における標的タンパク質量を検出します。酵素β-ガラクトシダーゼをEnzyme donor (enhanced ProLabel; ePL) とEnzyme acceptor (EA)とに2分割し、ePLと標的タンパク質の融合タンパク質を細胞内に発現導入します。この細胞にEAおよびβ-ガラクトシダーゼ基質を含む専用の検出用試薬を添加すると、ePLとEAは相補的に結合し活性型β-ガラクトシダーゼを形成するので、β-ガラクトシダーゼ基質の化学発光量シグナルを検出することにより標的タンパク質量を定量することが可能となります（Fig.1）。

細胞内における標的タンパク質量は、タンパク質の発現と分解のバランスにより、定常状態に保たれます。細胞膜を透過可能なリード化合物は、細胞内において標的タンパク質と結合することにより、標的タンパク質の半減期を変化させます（Fig.2）。細胞あたりのePL融合タンパク質量の増減を検出することにより、細胞内微小環境下におけるリード化合物の標的タンパク質への結合や作用を評価することが可能となります。
