What ELISpot and FluoroSpot reveal about these (not-so-secret) stars in the fight against SARS-CoV-2

The ELISpot method, originally developed to quantify the number of immunoglobulin-secreting cells, has evolved to be one of the most sensitive, high-throughput and commonly used techniques to monitor cell mediated immune responses by measuring the secretion of cytokines. It is used for studies of immune responses in a wide range of diseases, e.g. allergy, infections, autoimmune diseases and cancer. The IFN-g ELISpot assay is considered a standard tool to evaluate antigen-specific T cell immunity during the development of vaccine candidates and monitoring of new vaccines. Although ELISpot measures secretion of one cytokine, it can be used to measure secretion of up to four cytokines in one well with a slightly different approach called FluoroSpot, using fluorescent labels.

Although ELISpot measures the secretion of one cytokine, it can be used to measure secretion of up to four cytokines in one well with a slightly different approach called FluoroSpot, using fluorescent labels. FluoroSpot allows for analysis of polyfunctional T cells producing two or more cytokines simultaneously, and the relative amount of cytokine secreted per cell can also be determined. CD4+ T-helper subsets induced by natural infection or vaccination can be delineated using IFN-γ (Th1) / IL-5 (Th2) / IL-17A (Th17) / IL-22 (Th22) FluoroSpot. IFN-γ / IL-2 / TNF-α FluoroSpot can be used to quantify frequencies of central memory (TCM), effector memory (TEM) and terminally differentiated effector memory (TEMRA) T cells. CD8+ cytotoxic T-cell frequencies can be quantified using IFN-γ / GrzB FluoroSpot and divided into TCM and TEM/TEMRA populations

ELISpot and FluoroSpot assays have been used extensively to map SARS-CoV-2 specific T-cell responses induced by natural infection and to quantify T-cell responses after vaccination
