高い特異性と親和性！Nano-Secondary® アルパカ抗マウス免疫グロブリンG2b Alpaca anti-mouse IgG2b 抗体（Alexa Fluor® 647標識）

Alpaca anti mouse IgG2b 抗体（Nano-Secondary® alpaca anti-mouse IgG2b, recombinant VHH, Alexa Fluor® 647）は、マウスIgG2b（mouse IgG2b）を検出するAlexa Fluor® 647 標識済みの二次抗体です。

アルパカ（Alpaca）を宿主動物（免疫動物）として作製された「組換えVHHモノクローナル抗体」（VHH：ラクダ科動物の「重鎖抗体」由来の抗原結合ドメイン、Nanobody® としても知られます）であり、高い特異性と親和性を示します。抗体サイズが小さく、高浸透性および高解像度の免疫蛍光染色を可能にします。

■ VHH抗体とは?

VHH抗体は、Nanobody® としても知られるラクダ科動物の「重鎖抗体（Heavy chain-only antibody）」由来の抗原結合ドメインです。一般的な抗体と比べて、10分の1程度とサイズが小さく、高い特異性と親和性を示すことで知られます。

■ Nano-Secondary®とは?

Nano-Secondary®（ナノ二次抗体）は、蛍光色素とアルパカ由来のVHH抗体からなる、動物種/抗体サブタイプ特異的な二次抗体です。従来のモノクローナル抗体やポリクローナル抗体とは異なり、無血清発現系を用いた組換え抗体として安定的に産生されます。また、開発過程で他種由来IgGや血清タンパク質との交差反応性を検証したクローンを選別しており、予備吸着不要で高い特異性と低バックグラウンドを示します。

抗体サイズが小さいため、高浸透性かつ高解像なイメージングに寄与し、免疫蛍光染色（IF）を中心としたアプリケーションに利用できます。

また、Nano-Secondary®は、免疫蛍光染色（IF）やウェスタンブロット（WB）で『ワンステップ染色』（一次抗体と二次抗体（Nano-Secondary®）を同時にインキュベーションする方法）にも対応しています（参考プロトコール参照）。一価のVHH二次抗体は、小サイズでかつ高い特異性と親和性を示すため、従来の二価の二次抗体と異なり、一次抗体のエピトープに対するアクセス性や細胞内への浸透性を妨げる大きなクラスター（複合体）を形成しません。

Multiple Nano-Secondaries can be applied for multiplex fluorescent Western blotting. This allows multiple targets to be analyzed simultaneously on the same blot at the same time.
