GPCRはあらゆる疾患領域で極めて重要な治療標的分子であり、現行医薬品の約4割はGPCRに作用することで臨床効果を発揮すると言われています。一方、ヒトに存在する300種類以上のGPCR(感覚受容体除く)のうち、医薬品の標的とされているGPCRは一部に限られます。200種以上のGPCRで新薬開発が進んでいない理由の一つは、薬剤候補化合物を評価するためのアッセイ系が乏しく、薬理的解析が困難であることです。

タンソーバイオサイエンス社は、独自のレポーター細胞及びGαタンパク質を組み合わせることで、これまで適切な活性評価系のなかったGPCRを含め、ヒト全GPCRに対する高感度な機能解析系を構築しました。タンソーバイオサイエンス社GPCR委託試験は、幅広いGPCRに対するオンターゲット/オフターゲット探索、特定のGPCRに対するスクリーニングや最適化など、多様な目的でご利用頂けます。

原理

リガンドにより活性化されたGPCRは、共役するGタンパク質を介し複数の細胞内情報伝達系を活性化する。タンソーバイオサイエンス社のアッセイは、細胞内情報伝達系の下流で分岐したシグナルを再合流させることで、積算効果によりシグナルを増幅する仕組みをアッセイ用細胞に導入している。本技術は、レポーターであるルシフェラーゼ活性を指標に、標的GPCRに対するサンプルの刺激特異的な活性を鋭敏に評価することを可能にした。
