背景

■ HumanKine®（ヒューマンカイン）とは?

『HumanKine®（ヒューマンカイン）』は、ヒト細胞（HEK293細胞）発現系によって産生された組換えタンパク質製品です。大腸菌等の細菌（原核生物）発現系によって得られるタンパク質とは異なり、ヒト（真核生物／哺乳動物）由来細胞を用いて産生されるタンパク質は、ネイティブ型のコンフォメーションを取り、適切な翻訳後修飾を受けた「本物」のヒトタンパク質（'authentic' human proteins）として機能し、優れた生物活性と安定性を示します。ヒューマンカインは、アニマルフリーおよびゼノフリー条件下で、タグが付加されないタンパク質として産生されます。エンドトキシンフリーのタンパク質をキャリアフリー包装にてお届けします。

「cGMPグレード」組換えタンパク質も提供できます

HumanKine®（ヒューマンカイン）は「標準研究グレード」に加えて「cGMPグレード」製品として提供できます。いずれのグレードであっても、米国のcGMPグレード施設で同一の発現系、製造工程、作業手順によって製造されています。そのため、2種類のグレード間における品質面や性能面の違いは実質的にはありません。それぞれのグレード間での大きな違いは、ISO 13484認証およびcGMP準拠の厳格な品質プロセスに準拠しているかどうかです。cGMPグレード製品の場合、規制に沿った文書化、徹底したQC、トレーサビリティ対応を実施しています。

安定性と保存方法	Lyophilized proteins are stable for 1 year from the date of receipt if stored between (-20℃) and (-80℃). Upon reconstitution we recommend that the solution can be stored at (4℃) for short term or at (-20℃) to (-80℃) for long term. Repeated freeze thaw cycles should be avoided with reconstituted products.
再溶解	Briefly centrifuge the vial before opening. It is recommended to reconstitute the protein to 0.2 mg/mL in sterile 1x PBS pH 7.4. Gently swirl or tap vial to mix.

Purity of recombinant human Betacellulin was determined by SDS- polyacrylamide gel electrophoresis. The protein was resolved in an SDS- polyacrylamide gel in reducing and non-reducing conditions and stained using Coomassie blue.

Recombinant human Betacellulin (HZ-1339) stimulates dose-dependent proliferation of the NIH/3T3 mouse fibroblast cell line. Viable cell number was quantitatively assessed by Prestoblue Cell Viability Reagent. NIH/3T3 cells were serum starved during treatment with increasing concentrations of recombinant human Betacellulin for 72hrs in defined medium. The EC50 was determined using a 4- parameter non-linear regression model. The EC50 values range from 1 - 10ng/mL.
