要約: GTP(グアノシン三リン酸)アーゼであるRasのGTP結合型は、Rafキナーゼやホスファチジルイノシトール3キナーゼ(PI3K)などの多様なエフェクターに結合することによって、複数のシグナル伝達経路を開始させる。Rasの活性は、グアノシン二リン酸の放出とGTPの結合を促進するグアニンヌクレオチド交換因子によって増大し、またGTP加水分解を促進するGTPアーゼ活性化タンパク質によって抑制される。KRASは、がんにおいて最も頻繁に突然変異がみられるRAS遺伝子である。今回われわれは、野生型K-Rasのグアニンヌクレオチド結合モチーフ中のリジン-147がモノユビキチン化されることによって、GTP結合の亢進が起こりうることについて報告する。さらに、ユビキチン化によって、K-Rasの発がん性Gly12Val変異体の、下流エフェクターであるPI3KとRafへの結合が亢進した。したがって、モノユビキチン化はK-RasへのGTP結合を増強し、特異的な下流エフェクターへの親和性を増大させる可能性があり、未知のRas活性化機構をもたらすかもしれない。
