要約:プロテインキナーゼの触媒活性は、通常は翻訳後修飾と多様な組合せの調節タンパク質によって厳密に制御されている。プロテインキナーゼは極めて動的な酵素であり、様々な活性化状態におけるキナーゼの構造、および調節タンパク質との複合体の構造から、この大きくて多様なタンパク質ファミリーの複雑な調節機構について、きわめて重要な洞察が得られている。プロテインキナーゼA(PKA)の結晶構造は、キナーゼの触媒機能をわれわれが理解する基準モデルとなった。今回、20年以上後になって、完全長四量体PKAホロ酵素の高分解能モデルが発表され、PKAのアロステリック活性化の基盤となる構造機構が明らかになった。
