要約
自閉症スペクトラム症(ASD)は、複数の遺伝的危険因子と関連がある、高頻度でみられる神経発達症である。ヒト疾患関連の一塩基多型(SNP)のほとんどは遺伝子内ではなく遺伝子発現を調節する調節領域内に位置する。調節領域の機能はエピジェネティック(後成的)な機構によって決定される。発生の細胞学的基盤と長期記憶の形成とのあいだ、とくに両過程におけるエピジェネティックな機構の役割には、だいぶ前から類似点が認められている。われわれは、マウス海馬におけるクロマチンへの接近可能性が学習によって変化する様子を、新たな高処理量シーケンシングによるバイオインフォマティクス戦略によって解析した。われわれがDEScan(differential enrichment scan)と呼ぶこの戦略を用いることで、多重反復を含むエピゲノミクス実験から得られたデータの解析が可能になり、2,365調節領域(大半はプロモータ領域)におけるクロマチンへの接近可能性の変化が明らかになった。学習によって調節されるプロモータは、マウスの前脳形成中に活動的であり、かつbivalent(二価性)なプロモータであることを示すエピジェネティック修飾が高密度でみられた。これらのプロモータは、イントロンに偏在しており、記憶の固定中および想起中の遺伝子発現および選択的スプライシングと複雑な関係を示し、既知のASDリスク遺伝子に関連するデータセットに高頻度で認められた。このようなプロモータの1つであるSHANK3プロモータ6の臨床コホートにおける遺伝型決定によって、SNP rs6010065がASDと関連することが明らかにされた。今回のデータは、学習は発達をエピジェネティックに再現するという考えを支持し、マウスにおいて行動学的に誘導されたエピジェネティックな変化によって、患者の脳障害に関連する調節領域を浮き彫りにできることを実証するものである。
