生殖器がんを有する女性ではしばしば、プロゲステロン受容体(PGR)アイソフォームであるPGRAおよびPGRBの相対的存在量に相違が認められる。このようなPGRアイソフォーム比が生殖器の維持において持っている重要性を評価するため、われわれは、全てのPGR陽性組織においてPGRAまたはPGRBを過剰発現しているマウスを作製した。PGRA過剰発現マウスはほとんど生殖器腫瘍を発生しなかったが、PGRB過剰発現マウスではその全例に、卵巣黄体細胞に由来する卵巣新生物が発生した。PGRB過剰発現マウスの卵巣腫瘍のトランスクリプトーム解析から、AKTシグナル伝達の亢進と、ヒト卵巣がんおよび子宮内膜がんと類似した遺伝子発現シグネチャーが明らかにされた。PGRB過剰発現マウスにPGRアンタゴニストであるRU486を投与したところ、腫瘍増殖が停止し、細胞周期関連遺伝子の発現が低下した。このことは、腫瘍増殖および細胞増殖がアイソフォーム依存性に加えてホルモン依存性であることを示していた。PGRBシストロームの解析から、分裂期開始の重要な調節因子であるタンパク質をコードしている遺伝子での、結合イベントが特定された。本研究は、in vivoにおけるPGRA存在量に比べたPGRB存在量の増加が細胞周期回転を刺激することで、腫瘍形成を誘導するという機構を示唆している。
