要約:T細胞免疫グロブリン・ムチンドメイン(T cell immunoglobulin and mucin domain:TIM)タンパク質は、T細胞においては細胞表面シグナル伝達受容体であり、抗原提示細胞や腎尿細管上皮ではスカベンジャー受容体である。今回われわれは、TIMタンパク質がNUR77(アポトーシスと細胞の生存に関わる核内受容体)の分解の媒介において機能していることを明らかにした。TIMタンパク質はホスホイノシチド3キナーゼ依存性の経路でNUR77と相互作用し、NUR77のリソソームでの分解を媒介していた。またわれわれは、TIM-1がクラスリン依存性の構成的なエンドサイトーシスによって、細胞表面との間で動的に循環していることも明らかにした。この過程を遮断したり、TIM-1中のホスファチジルセリン結合ポケットを変異させたりすると、TIM-1が媒介するNUR77の分解が抑制された。in vitroの腎障害モデルでは、TIM-1のサイレンシングによってNUR77量が増大し、上皮細胞の生存率が低下した。これらの結果は、TIMタンパク質が免疫細胞の機能および障害に対する腎臓の応答に影響を及ぼす可能性を示している。
