要約 Apoer2は、アポリポタンパク質ApoEに結合する、中枢神経系に必須の受容体である。Apoer2のさまざまなスプライスバリアントが産生される。われわれは、O結合型糖鎖(OLS)ドメインをコードするエクソン16を欠損したApoer2が、細胞においてApoer2のタンパク分解性プロセシングと存在量を変化させ、マウスにおいてはシナプスの数と機能を変化させることを示した。このスプライスバリアントを発現する培養細胞においては、OLS欠損Apoer2の細胞外切断が減少し、結果として、Apoer2の細胞内ドメインのγ-セクレターゼ依存性放出が阻害された。OLSドメインを欠損したApoer2を発現するマウスでは、脳内のApoer2存在量、海馬の棘突起密度、グルタミン酸受容体の存在量が増加したが、シナプス効率が低下した。OLSドメインを欠損し、グルタミン酸受容体シグナル伝達を促進する選択的にスプライスされた細胞質尾部領域を含む型のApoer2を発現するマウスでは、学習と記憶に関連する現象である、海馬の長期増強(LTP)の亢進が認められた。しかし、これらのマウスでは、モリス水迷路における空間学習の増強は認められず、手がかり恐怖条件付け(cued fear conditioning)が減少した。変異タンパク質の量が野生型マウスのApoer2量と同程度になるように、変異Apoer2アレルの発現を低下させると、棘突起密度、海馬LTP、手がかり恐怖学習が正常化した。これらの結果から、ApoE受容体の、シナプスグルタミン酸受容体活性の調節因子として役割が証明され、差次的な受容体グリコシル化が、シナプス機能および記憶の調節因子の候補として確定された。
