EML4は、微小管の安定性を高める微小管結合タンパク質である。われわれは、細胞周期中のEML4の調節について調べ、間期にはEML4は微小管格子に沿って複数の焦点をなすように点状に分布しているが、分裂期には紡錘状の微小管との結合が減少することを見出した。また、微小管沈降法とクライオ電子顕微鏡による3次元再構成によって、EML4の塩基性N末端ドメインが微小管表面のαチューブリンおよびβチューブリンの酸性C末端尾部へのEML4の結合を媒介していることを明らかにした。有糸分裂期キナーゼNEK6、NEK7は、in vitroでEML4のN末端ドメインのSer144、Ser146をリン酸化し、細胞中の両キナーゼを欠損させると、有糸分裂微小管に結合するEML4の量が増加した。S144A-S146A二重変異体は、有糸分裂微小管と不適切に結合するだけでなく、その安定性を高め、染色体の集合を妨げた。さらに、NEK6またはNEK7を恒常的に活性化させると、EML4と間期微小管との結合が減少した。まとめると、これらのデータは、NEK6およびNEK7に依存したリン酸化によって、染色体の効率的な集合に必要な有糸分裂微小管からのEML4の解離が促進されるというモデルを支持するものである。
