間質相互作用分子1(STIM1)には、2つの重要な機能がある。小胞体内のCa2+センシングと、細胞膜Ca2+流入を可能にするストア作動性Ca2+チャネルOrai1の活性化である。分子動力学(MD)シミュレーションと生細胞記録を組み合わせ、活性化時の内腔STIM1ドメインの連続的なCa2+依存的コンフォメーションを決定した。さらに、複数のCa2+イオンに結合できる標準および非標準EFハンドドメイン内の残基を特定した。MDシミュレーションでは、単一のCa2+イオンで、内腔STIM1複合体を安定化するのに十分であった。Ca2+ストアの枯渇により、2つのEFハンドが不安定になり、安定したSAMドメインと一緒に形成される疎水性溝の解消が引き起こされた。標準的なEFハンドおよび疎水性溝を標的とした尿細管凝集性ミオパシーまたはがんに関連する点突然変異は、恒常的にクラスター化されたSTIM1を生成し、それは、Orai1チャネルを介したCa2+流入の活性化に関連していた。われわれは、これらの結果に基づき、STIM1 Ca2+結合モデルを提示し、分子レベルでのSTIM1活性化の現在知られている初期ステップをより正確に示した。
