要約:Raf-MEK(マイトジェン活性化または細胞外シグナル調節プロテインキナーゼキナーゼ)-ERK(細胞外シグナル調節キナーゼ)から成るマイトジェン活性化プロテインキナーゼ経路などの三段階キナーゼモジュールは生物において広く見られることから、この構造が進化的に有利な特性を伝えると考えられる。本研究では、三段階キナーゼ増幅因子モジュールと負のフィードバックの組合せが、電子回路に用いられ、頑強性の付与、出力安定化および非線形シグナル増幅の線形化を行う負帰還増幅器(NFA)のデザイン原理の要点を再現していることを示す。数学的モデリングおよび実験的検証を用いて、ERK経路が(i)内因性スイッチ様活性化動態を段階的線形応答へと変換し、(ii)NFAモジュール内の反応速度の変化に頑強性を与え、(iii)薬物による増幅因子のかく乱に応じて出力を安定化するというNFAの特性を有することを示した。このような特性が活性化動態および薬物応答性などの生物学的行為を決定する。
