要約
メラトニンは夜間に生成され、睡眠と概日リズムを制御している。Gタンパク質共役受容体(GPCR)の1種であるメラトニン受容体MT2をコードしているMTNR1Bの機能喪失バリアントは、2型糖尿病(T2D)のリスク増加と関連している。特異的なT2D関連シグナル伝達経路を同定するため、われわれは複数のシグナル伝達エフェクターの自発性(リガンド非依存性)およびメラトニン誘導性の活性化をモニタリングすることで、40種のMT2バリアントのシグナル伝達アウトプットをプロファイリングした。遺伝子相関解析から、メラトニン誘導性のGαi1およびGαzタンパク質活性化およびMT2への自発的なβ-アレスチン2結合の欠如は、T2Dリスク増加と統計学的に最も有意に相関することが明らかとなった。in silico進化系統解析により計算的に予測されたバリアントの影響は、各バリアントに測定された表現型への影響と強く相関していることが示され、将来的なGPCRバリアント研究のための予測ツールが提供された。まとめると、今回の大規模な機能研究から、ヒト集団に存在する複数のGPCRバリアントのポストゲノム解析に使用できる枠組みが提供された。T2Dリスクと、シグナル伝達経路特異的な欠陥の相関は、経路特異的な個別化治療的介入のための道を開き、またこれにより、メラトニンを検出できないが受容体の自発的活性化が生じている日中における、MT2機能の潜在的意義が解明された。
