インバリアントナチュラルキラーT(iNKT)細胞は、CD1d分子上で提示されている糖脂質抗原を認識するインバリアントT細胞受容体(TCR)を発現している、自然免疫様Tリンパ球である。これらの細胞は表現型としても機能的にも通常型T細胞と異なっている。われわれはiNKT細胞の代謝活性を特性評価し、その活性型の表現型に一致して、ナイーブな通常型CD4+T細胞と比べミトコンドリア呼吸活性が非常に低下しているが、解糖活性は亢進していることを見出した。iNKT細胞ではTCR会合の後、インターフェロン-γ(IFN-γ)の発現に重要な好気的解糖がさらに亢進した。解糖代謝はヘキソキナーゼ-IIのミトコンドリアへの移行および哺乳類ラパマイシン標的タンパク質複合体2(mTORC2)の活性化を促進した。解糖系の阻害はAktおよびPKCθの活性を低下させ、これが免疫シナプス内のTCRの再利用と蓄積を阻害した。免疫シナプスにおけるTCR蓄積の低下により、近位および遠位のTCRシグナル伝達経路の活性化、並びに活性化iNKT細胞におけるIFN-γの生成が減少した。われわれの研究は、解糖代謝がTCR再利用を増加させることでiNKT細胞のTCRシグナル伝達持続時間を延長し、IFN-γの生成を増強させることを実証している。
