要約:ナチュラルキラー(NK)細胞は、その活性化が活性化受容体と抑制性受容体からのシグナルの相互作用によって厳密に調節されている免疫系のエフェクター細胞である。活性化受容体からのシグナルはグアニンヌクレオチド交換因子Vav1のリン酸化を誘導するのに対して、抑制性受容体からのシグナルはSrcホモロジー2ドメイン含有プロテインチロシンホスファターゼ1(SHP-1)によるVav1の脱リン酸化を引き起こす。本研究では、数学的モデリングとNK細胞を用いた実験によって、分子レベルでの正のシグナルと負のシグナルの統合に関する洞察を得た。本研究のデータから、Vav1のリン酸化にはスイッチ様の調節が働き、その程度はNK細胞の細胞傷害活性と相関することが示された。このような実験結果を72個の数学的モデルから導いた予測と比較して、細胞傷害応答の発生にはSrcファミリーキナーゼとNK細胞上の活性化受容体との物理的会合が必須であることが示された。われわれのデータは、NK細胞の細胞傷害活性の決定におけるVav1の中心的役割を裏付け、リンパ球活性化の際の正のシグナルと負のシグナルの統合の分子的機構に関する洞察を与えるものである。
