要約:Akt1のリン酸化と活性化は、脂肪生成を促進する重要なシグナル伝達イベントである。しかし、Thr308およびSer473部位でのリン酸化を介したAkt1活性化を導く複雑な多段階プロセスも、Akt1が脂肪生成を刺激する機構も、完全には理解されていない。われわれは、BSD domain-containing signal transducer and Akt interactor(BSTA)がヒトおよびマウスのさまざまな細胞においてSer473部位でのAkt1リン酸化を促進することを確認し、BSTA-Akt1複合体形成におけるBSDドメインの働きを明らかにした。mammalian target of rapamycin complex 2(mTORC2)がBSTAのリン酸化ならびにAkt1との会合を促進し、このBSTA-Akt1の相互作用が、増殖因子刺激に反応したTORC2とAkt1の会合およびSer473部位でのAkt1のリン酸化を促していた。さらにbsta遺伝子をトラップしたマウス胚性幹細胞の解析から、脂肪細胞の分化促進において、BSTA並びにSer473部位でのAkt1リン酸化が必須の機能を果たしていること、また転写因子FoxC2をコードする遺伝子の発現の抑制が必要なことが明らかとなった。これらの知見は、BSTAがSer473部位でのAkt1リン酸化を促進する分子スイッチであることを示唆し、さらに、脂肪細胞の分化に重要なmTORC2-BSTA-Akt1-FoxC2を介するシグナル伝達機構の存在を明らかにした。
